The relationship between moisture, sucrose, laevulose, and dextrose during honey formation was studied in Apis mellifera L. from March 2022 to August 2023 in Chintamani, Chickaballapur district. Five stages-floral nectar, forager crop sac, house bee crop sac, unsealed honey cells, and sealed honey cells—were analysed. Moisture content decreased from 79.08% in floral nectar to 18.7% in sealed honey cells. Sucrose reduced from 20.24% to 4.21%, while laevulose and dextrose increased from 1.3% to 40.2%. The most significant moisture reduction occurred in unsealed honey cells (65%), followed by the house bee crop sac, forager crop sac, and sealed honey cells. Laevulose and dextrose showed similar trends, with the highest increases in unsealed honey cells (55% and 47%, respectively). The study indicates a correlation between declining moisture and sucrose levels and rising laevulose and dextrose levels, due to moisture utilization in sugar formation and the action of invertase and diastase enzymes from the honeybee hypopharyngeal and mandibular glands. Fanning behaviour of honeybees also plays a crucial role in honey elaboration. Data were subjected to CRD-factorial analysis and significant levels were determined at 5% (P <0.05) and 1% (P <0.01).
