Abstract

Mesalamine, also known as 5-aminosalicylic acid (5-ASA), is an anti-inflammatory drug used to treat inflammatory bowel disease (IBD). The primary goal of this study is to develop and evaluate a nanoparticulate drug delivery system using mesalamine incorporated into polymeric micelle-filled pulsincaps to treat IBD. The solubility comparison between the pure drug and the polymeric micelles showed that mesalamine in polymeric micelles had better solubility in both water and phosphate buffer at pH 6.8. FTIR and DSC analyses confirmed no interaction between the drug and excipients. Critical Micelle Concentration studies were conducted, and various polymeric micelle formulations containing 250 mg of the drug were prepared and evaluated for various different parameters. The optimized polymeric micelle solution was freeze-dried into a free-flowing powder and assessed for compatibility, scanning electron microscopy, and flow properties. Pulsincaps were prepared by coating the capsule body with ethyl cellulose and the cap with the enteric coating material Eudragit S100. The free-flowing polymeric micelle powder was filled into the capsule bodies, sealed with HPMC K100M. Dissolution studies showed the %CDR from the mesalamine-loaded polymeric micelle-filled pulsincaps was 91.63±0.10% after 12 hours. Stability studies of the polymeric micelles were conducted according to ICH guidelines.
