In vitro assessment of Plumeria pudica for antidiabetic properties using antioxidant assays and studies on 3T3-L1, β-TC6, and C2C12 cell lines

Abstract

Background: Diabetes mellitus is an abnormal glucose metabolic condition, characterized by reduced glucose transport from the systemic circulation to the tissues due to lack of insulin. In India Plumeria pudica plant is called naga champa which has been extensively utilized for its medical benefits, encompassing its antimicrobial, anti-inflammatory, anti-rheumatic, anti-ulcer, and anti-allergic qualities. Aim: In the present investigation, the ethanolic leave extract is used to evaluate in vitro anti-oxidant & antidiabetic activity in β-TC6 cells, 3T3- L1, and C2C12 cell lines. Methods: The phytochemical analysis of selected plant extracts has been done for confirmation of various constituents. The DPPH free radical scavenging activity was performed for antioxidant activity. The Insulin secretion and glucose uptake was determined in various cell lines. The Metformin & Glibenclamide are used as standard drug. Results: The phytochemical analysis of EEPP contains phenols and flavonoids. The IC50 value of test and standard is found to be 42.21 &11.27 respectively. The Insulin secretion was shown significantly (p<0.05) raised in β-TC6 cells, and glucose uptake significantly increased (p<0.01) in 3T3L1 cells, and C2C12 cell lines. Conclusions:- Presence of polyphenols, and flavonoids in test attributes antioxidant as well as antidiabetic effect and was confirmed by cell line studies.
