Syringaldehyde demonstrates analgesic, anti-nociceptive and antihyperalgesic potency in rodent pain models

Abstract

Discovery of novel treatments for management of various pain conditions is an unmet need. Syringaldehyde is a phytochemical compound with antiinflammatory, anti-oxidant potential. In this study, experimental animal models of analgesia, nociception and inflammatory pain were utilized to ascertain pharmacological potency of Syringaldehyde. Syringaldehyde was tested in mouse tail flick test and formalin-induced nociceptive pain. To assess the pharmacological effect of Syringaldehyde, mouse models of tail flick test and formalin- induced nociception were used. To assess effect of Syringaldehyde in inflammatory pain, Carrageenan induced thermal pain hypersensitivity (hyperalgesia) and complete Freund’s adjuvant (CFA) induced mechanical pain hypersensitivity (hyperalgesia) in rats was used along with analysis of expression levels of inflammatory mediators. Dose-dependent analgesic effect was seen with Syringaldehyde treatment in tail flick test, while Syringaldehyde pretreatment substantially reduced formalin-induced nociceptive pain in second phase of formalin test. In inflammatory pain induced with intraplantar injection of Carrageenan and CFA, Syringaldehyde treatment reversed thermal and mechanical hyperalgesia with inhibition of elevated inflammatory pain mediator levels. In this study, Syringaldehyde demonstrated analgesic, antinociceptive activity and inhibited inflammatory pain through modulation of inflammatory mediators.
