The present study is aimed to develop, optimize and evaluate polyherbal transdermal patch consists of boswellic acids, curcuminoids, diosgenin and rutin for treatment of arthritis. Traditionally, these phytochemicals are used in the treatment of arthritis. Matrix type of transdermal patch was formulated by solvent casting method using the combinations of polymers, Eudragit RL 100 and Eudragit RS 100, polyethylene glycol 400 as a plasticizer and l-menthol as a permeation enhancer. A 23 full factorial design was applied for the optimization of process variables such as polymers and penetration enhancer concentration. Selected responses were tensile strength and Q24. Patches were evaluated for physicochemical properties like thickness, weight variation, folding endurance, percentage flatness, percentage moisture content, percentage moisture uptake, water vapor transmission rate, drug content, in-vitro diffusion studies and ex-vivo skin permeation studies. All the formulations had desirable physicochemical properties. Formulation P8 showed maximum Q24, So P8 batch was studied further for ex-vivo skin permeation. Kinetic modelling suggested that non-Fickian type of diffusion is dominant mechanism for drug release. No erythema or edema was observed in the skin irritation test confirming the patch was non-toxic and safe. Stability studies were carried out according to ICH guidelines which clearly revealed that the optimized patch is stable. The formulated transdermal patch could release significant amounts of drugs into the systemic circulation. Polyherbal patch could provide an alternative to traditional NSAIDs for arthritis treatment.
