Exploring the therapeutic potential of Tinospora Cordifolia (Guduchi) against Indomethacin-induced ulcerative colitis in experimental animals: A preclinical study

Abstract

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by inflammation and ulceration of the colonic mucosa. Indomethacin, a nonsteroidal anti-inflammatory drug (NSAID), is commonly used to induce UC-like symptoms in experimental models. Tinospora cordifolia (Guduchi), a well-known medicinal herb, possesses anti-inflammatory, antioxidant, and immunomodulatory properties, making it a promising candidate for managing inflammatory disorders. This study aimed to evaluate the therapeutic efficacy of Tinospora cordifolia in indomethacin-induced ulcerative colitis in rats. UC was induced using indomethacin, and the effects of Tinospora cordifolia were assessed by monitoring body weight, inflammatory markers, haematological parameters, and histopathological changes in colonic tissue. Indomethacin-treated rats exhibited significant weight loss, elevated ESR and CRP levels, and altered haematological indices, including reduced Hb and RBC counts and increased WBC and platelet counts. Histopathological analysis revealed severe mucosal damage, ulceration, and inflammatory infiltration. Treatment with Tinospora cordifolia significantly improved body weight, reduced ESR and CRP levels, normalized haematological parameters, and restored colonic tissue architecture, promoting mucosal healing. These findings suggest that Tinospora cordifolia exhibits significant therapeutic potential in managing indomethacin-induced ulcerative colitis through its anti-inflammatory, antioxidant, and immunomodulatory mechanisms.
