Glimepiride (GL) is an oral hypoglycaemic agent with compromised solubility. The present work is intended to improve the solubility of this drug by formation of binary eutectic mixtures. The eutectic mixtures were synthesised using gallic acid, ascorbic acid, thiourea and caffeine as coformers as indicated by a significant depression in the melting points in the DSC analysis, accompani ed by no apparent change observed in PX RD analysis. The morphological differences were analysed by SEM analysis. A maximum of 14 and 63 times increase in solubility was observed in 0.1 N HCl and phosphate buffer pH 6.8 respectively in GL GA. Pharmacokineti c studies were performed on all the prepared eutectic formulations and compared with pure GL. GL GA provided highest plasma blood concentration of GL. As projected, the invitro data very well corroborate with in vivo results and the drug release was found to be maximum when an equimolar concentration of gallic acid was used as conformer, making gallic acid an ideal candidate for the cocrystal formulation. The research work presented a complete preliminary data on improved physicochemical properties of glime piride.
