Crohn's disease (CD), a subtype of inflammatory bowel disease (IBD), is characterized by chronic inflammation of the gastrointestinal tract, affecting various segments from the oral cavity to the perianal region. The disease significantly impacts patients' quality of life, and its prevalence has risen in recent decades. Despite advancements in treatment, current therapies are not curative and often have limitations. This study explores the potential of conditioned media (CM) derived from human umbilical vein endothelial cells (HUVECs) as a novel therapeutic approach for CD. In this research, CD was induced in adult male Wistar rats using indomethacin, followed by treatment with HUVEC-derived conditioned media (H-CM). Histopathological analysis of the colon and small intestine demonstrated that H-CM treatment alleviated chronic lesions, restored tissue structure, and improved mucosal architecture compared to the untreated group. Cytokine analysis further confirmed that H-CM treatment significantly reduced levels of pro-inflammatory cytokines, including TNF-α, IL-1β, and IL-6. These findings indicate that H-CM has beneficial effects in mitigating the immunopathological changes associated with CD and could serve as a promising alternative to traditional cell-based therapies. The study provides evidence that endothelial cell-derived CM holds therapeutic potential for managing CD, offering a safer and more accessible treatment option.
