In vitro shoot regeneration from immature explants of Zea Mays L.

Abstract

In maize tissue culture, immature embryos (IEs) serve as highly regenerative explants ideal for achieving high plantlet regeneration. This study reported how explants size and hormone concentrations influence key in vitro responses, including callus induction, and plantlet regeneration. Immature embryos of various size categories from maize lines (VNR 4352) were cultured on MS media containing varying levels of 2,4-D and 8 μM/l Kinetin for callus development. 89.38 % of calli were obtained on MS medium containing combination of auxin and cytokinin in MS media produced 89.38% embryogenic calli. Maximum regeneration (70%) was observed when embryogenic calli were placed on MS medium supplemented with 30 gL-1 sucrose and 3μM/l BAPfour week.a maximum number of shoots (4.0 ±2.13) and the highest shoot length (4.34 ± 0.62) was achievedwithin 4 weeks of culture. Furthermore shoot elongationwas achieved on medium containing 4.0 μM/l BAP. The elongated micro shoots were rooted on MS medium supplemented with 2.0 μM/l indole-3-butyric acid. The regenerated plantlets with roots weresuccessfully hardened on earthen pots after proper acclimatization under greenhouse conditions.Embryos of 3.0 mm size produced optimal results. The study confirmed that embryogenic calli obtained from immature embryo of maize is fully capable to regenerate into whole plant under Optimized MSMedium and cultural conditions.
