TNF-α, TGF-β and INF-γ are cytokines involved in the pathogenesis of various tumors, including nasopharyngeal carcinoma (NPC). Many research have examined the link between polymorphisms of these cytokines and NPC, the results remain uncertain. Our study aims to determine whether there is a link between genetic polymorphisms of TNF-α-308G/A (rs1800629), TGF-β1 codon 10T/C (rs1800470) and codon 25G/C (rs1800471) and INF-γT/A (rs2430561), by PCR-SSP method using DNA from algerian patients, and the onset of the desease. It also aims to identify minor alleles associated with this pathology. In our study, cytokine gene polymorphisms showed no association with NPC, genotypic frequencies being non-significant (P > 0.05). However, several specific alleles were significantly associated with NPC, such as the G allele of TNF-α (P = 0.043), the G allele of TGF-β1 at codon 25 (P = 0.021), and the T allele of INF-γ (P = 0.043). These results suggest a possible involvement of these alleles in NPC in an Algerian population, their effect ramains modest and requires cautions interpretation, in the absence of confirmation by genotype frequencies. Genetic similarity analysis via a Heatmap examined by R software, revealed that most patients share homozygous genotypes for a specific polymorphism, as well as common alleles.
