Surface Sterilization Methods for Explant in DifferentVarieties of Potato (Solanum tuberosum)

Abstract

The process of surface sterilization is a critical step in the development of explants that are both healthy and viable for the purpose of tissue culture applications. Mostof surface impurities may be eradicated by the process of surface sterilization using an appropriate sterilizing agent. The purpose of this research was to provide a technique of disinfection that is efficient for the regeneration of shoots of Solanum tuberosum by using apical buds. A total of nine treatments were conducted for the sterilization of the apical bud explant of ten different varieties of potatoviz. Kufri Uday, Kufri Chipsona-1, Kufri Jyoti, Kufri Pukhraj, Kufri Surya, Kufri Lalima, Kufri Lima, Kufri Khyati, Kufri Mohan and Diamond were used. Apical bud is used as an explant and it was kept for several time periods in the sterilizing agents, with each treatment repeated three times. It incorporates Tween- 20, Bavistin, Ethyl alcohol, and Mercuric Chloride. The explants were cleaned with distilled water for 3-4 times after every step of sterilization. The findings showed that the treatment T8(Bavistin 0.3% (30 minutes) + Ethyl alcohol 70% (0.5 minutes) + 0.1% Mercuric chloride (1 minute)) had the least contamination and the highest survival rate and least browning and lowest contamination among survived explants of all the 10 varieties, followed by the T7(Bavistin 0.3% (30 minutes) + Ethyl alcohol 70% (0.5 minutes) + 0.1% Mercuric chloride (0.5 minutes)) andT5 (Bavistin 0.3% (30 minutes) + 0.1% Mercuric chloride (0.5 minutes)) treatments. However, in treatments T1(Bavistin 0.3% (25 minutes) + Ethanol 70% (0.5 minutes) + Mercuric chloride 0.1% (10 minutes)) and T2(Bavistin 0.3% (25 minutes) + Ethanol 70% (0.5 minutes) + Mercuric chloride 0.1% (5 minutes)) contamination was found least but in explants browning has started after a week, that leads to the death of explants, it was due to longer exposure to mercuric chloride i.e.10 minutes and 5 minutes.It is essential to pay attention to the time while cultivating potato tissue via the apical bud.
